The Hammers are enjoying an impressive Premier League season and will be targeting a run in the FA Cup in their last year before moving to the Olympic Stadium.
West Ham could hand a start to Dimitri Payet after he made his return from injury as a substitute in last weekend's win over Liverpool.
Wolves come into the cup tie in decent form after winning their last three matches to move up to 11th in the Championship.
It is the first meeting of the sides since 2011 when the Hammers won 2-0 in the Premier League at Upton Park.
